Michigan Bans NCAA Schools From Cyberstalking Student-Athletes

Michigan has become the latest state to ban schools from requiring their student-athletes to register their personal email and social media credentials.? Michigan joins Delaware, California, and New Jersey in banning NCAA schools from requiring their students to verify their personal digital accounts in order to apply or attend school, keep their scholarships, or participate in intercollegiate athletics.

Michigan's legislation is the most comprehensive in the country because it also bans elementary, middle, and high schools from also requiring their students to turn over their personal digital account information.? In a nutshell, the new law generally bans all schools from requesting their students provide them access to their personal social media/digital media usernames, passwords and/or content.? This policy affirms that the state of Michigan will not allow its schools to act like China who is requiring its citizens to register their personal digital accounts so the government may "monitor" everything their citizens do online.

Michigan's legislation may save Michigan schools tens of millions of dollars per year that may have been utilized to contract with companies that offer cyberstalking services to track the digital activities of students, their families, and friends.? The companies that sell cyberstalking software to schools use terms like, "monitoring", "educating", and "leading" when describing their services, and/or companies.? In addition, if you perform due diligence on the founders of the companies that offer these so called "monitoring" or "educating" services you may notice they have no verifiable professional credentials that demonstrate that any sports (college, amateur, or professional) organization should engage them for social media or education related services.

Some of these companies are also stating that they support social media privacy legislation which if true means they support a ban on their cyberstalking services.? In order for any social media "monitoring" (cyberstalking) software to properly work it needs a student to verify his personal digital credentials.? Absent student verification these services may be monitoring fake digital accounts.
???
Any public school that engages a firm to "monitor" (cyberstalk) their students online may in the near future receive a letter from their state's attorney general, or the U.S. Department Education, or the U.S. Federal Trade Commission, or a law firm regarding their practices.? Schools that? "monitor" (cyberstalk) their students online may soon encounter steep fines, lawsuits, or a loss of education funding that may amount to tens of millions of dollars. ?

The bottom line is that public schools that engage self-described "social media experts"/"social media education & monitoring services"/"social media protectors of reputation" may create tremendous personal safety and privacy problems for their student-athletes, and massive legal liability issues for their institutions and taxpayers.

Remains of Conn. gunman claimed for burial

(AP) ? The body of the man who killed 26 people at a Connecticut elementary school has been claimed for burial.

Connecticut Medical Examiner Wayne H. Carver II tells the Hartford Courant (http://cour.at/TrQFyH) that the remains of Adam Lanza were claimed several days ago by someone who wanted to remain anonymous.

A spokeswoman at Carver's office told The Associated Press she could not release details about the status of Lanza's remains.

The 20-year-old Lanza killed 20 first-graders and six educators at the Sandy Hook Elementary School on Dec. 14. He also killed his mother in their Newtown home before going on the rampage and then committing suicide.

A private funeral was held earlier this month in New Hampshire for his mother, Nancy Lanza.

Police have not offered a motive for the killings.

Jan Detlefsen commented on Terence Gannon's blog post Interesting UAV Work Going on at the University of Calgary

Over the last few of months I have been making regular visits to the lab of Dr. Alejandro Ramirez-Serrano?at the University of Calgary. ?Called the?Autonomous Reconfigurable/Robotic Systems Laboratory (AR2SL) it specializes in research related to (from the web site): 1) Ground and aerial?unconventional unmanned vehicle systems (UVS), 2)?reconfigurable intelligent mobile robotic systems, 3)?swarm robotics, 4)?intelligent control for UVS, and 5)?search & rescue applications of mobile robotics.

Roughly once a month, and on a very informal basis, a tour is organized where there is a walkthrough of the lab followed by lunch and discussion. ?The objectives are simple: expose Dr. Ramirez-Serrano's work to a broader audience, and try and coalesce interest in UAV work in Southern Alberta. ?Thought is also being given to setting up a semi-permanent Meetup group to pursue these objectives.

If you would like to be included in one of these upcoming tours, by all means, please get in touch and we can make the necessary arrangements. Storm rakes half of nation; Arkansas still dark

LITTLE ROCK, Ark. (AP) ? As a the muted ends of a powerful winter storm that has killed more than a dozen people plodded through the Northeast, many in Arkansas were seeking warmth and shelter against the cold prospect of life without electricity into the new year.

A Christmas Day blizzard dumped more than 15 inches of snow on the state, causing massive damage to power lines that have affected more than 200,000 customers.

With the bleak word from the state's largest utility that the lights could be out until after the start of the new year, many residents who awoke snowbound Wednesday morning found themselves jamming the city's hotel rooms by Thursday night.

"I'm coping with hot toddies and peanuts," said Lynda Johnson, who lined up a series of hotel stays through hotels.com to make it at least through Saturday night. She has already been to the movies ? she saw "Django Unchained" ? and checked in with neighbors multiple times to see if the lights are back on.

They aren't.

Deena Brazell spent a night in her car for warmth, though she hadn't planned it that way.

"Everything in the apartment is electric. I stayed in the apartment the first night. After that, it got cold really quick," she said. "I went out to charge the phone and fell asleep, then I just decided to stay."

After the storm's peak early Wednesday, homes and businesses from border-to-border had lost power. Johnson, and several others, said they were hoping the power would be back on Wednesday after spending Christmas night in the dark. Butut then the president of the state's largest utility announced that some of the outages would persist at least into New Year's Day. Little Rock was among the cities hardest hit.

"We spent the first night at home and turned on the fireplace, but it doesn't give off a lot of heat," said Kathy Garner, who sought refuge at her sister and brother-in-law's house in Maumelle, a Little Rock suburb.

In a typical year, tornadoes bring Arkansas' worst weather, but the damage is isolated and linemen have a relatively easy time fixing the power grid.

This week's storm was epic by comparison, and despite the jokes ? "In Wisconsin, we call this Tuesday" ? as of Thursday night there was more snow on the ground in Little Rock than Milwaukee.

"You run out of money fast," Johnson said. "The things you had planned to do, you can't do. You need food, clothing and shelter. Since I'm not home, I have to find someplace for shelter. Then you have to find something to eat."

The storm system responsible for the misery roared out of the Rockies early Tuesday with blizzard conditions in southwestern Oklahoma and tornadoes along the Gulf Coast.

After sweeping across Arkansas, giving Little Rock its first white Christmas since 1926, it rolled into the Midwest and Northeast before moving on to Canada. Up to 20 inches of snow fell in the Adirondacks of New York; Indianapolis had 7.5 inches, its greatest snowfall in four years; and 4-6 inches fell in and around Concord, N.H.

"I'm going to be shoveling all day, just trying to keep up with the snow, which is impossible," said Dale Lamprey, clearing the sidewalk outside the legislative office building near the New Hampshire Statehouse.

Nationwide, at least 17 people died because of the ice, snow and wind. Deaths from wind-toppled trees also were reported in Texas and Louisiana, but car crashes caused most of the fatalities.

A Michigan woman who was riding in a car that struck a tree and two people riding in a car that slid across the center line of a road in Arkansas and hit another vehicle.

Two people were killed in Kentucky crashes, a New York man was killed after his pickup truck skidded on an icy road in northwest Pennsylvania, and an Ohio teenager died after losing control of her car and smashing into an oncoming snowplow.

Forty-two students traveling to London and Dublin were stuck in the Nashville, Tenn., airport thanks to poor weather in the Northeast. The frustrated students, from universities in Tennessee and Kentucky, were supposed to leave Wednesday and arrive in London on Thursday.

"It's a two-week program, so it's shortened already," said Joe Woolley, spokesman for the Cooperative Center for Study Abroad.

Farther east, the storm knocked out power to more than 7,000 homes and businesses in Maryland. In New Jersey, gusts of more than 70 mph were recorded along the coast, and the weather service issued a flood warning for some coastal areas. There were about 800 power outages in Vermont, but only a handful in neighboring New Hampshire.

Back in Arkansas, utility workers struggling in freezing temperatures restored power to nearly a third of their customers that lost power during the Christmas storm, but that still meant that more than 135,000 homes and businesses were in the dark as forecasters predicted another round of freezing rain for Friday.

"You just want to be home," Garner said at her sister's house. "You just want to be in your own bed. There's nothing like the comfort of your own home."

Basic question about Windows 8 UI - Computers, Math, Science ...

Its very clear that Microsoft is trying to beat Apple to the Marriage of mobile and personal computing. The problem is I think their execution is not working relatively well. I agree that Win8 on a personal computing device looks to behave like a mobile device, and this causes en masse confusion. While I applaud Microsoft for their attempt to innovate something unique, I just dont think its working terribly well.

I would feel better, and I think a few others would say the same thing, if Microsoft were to focus on under the hood innovation (this made them known for things like Exchange and Active Directory, and although Active Directory is a real bastardization of LDAP and nothing new, it was something that made them popular). A total Security Redesign, the elimination of ActiveX and DCOM controls, a more decentralized order of operation (Im sure that MS could re-aquire the rights to Xenix its own version of Unix back in the day....yes....Microsoft owned a unix variant at one time, make no mistake about that), and overall stability improvements.

As it stands now constant redesigns to the desktop across all platforms that care about it are confusing users more and more. Microsoft redesigns, and it loses people (and Win8 may be the very release that drives users away from it and to alternatives like Apple and Unix variants -- Linux included). Apple is slowly trying to marry without actually doing it, the mobile device and the personal computing device, but without changing the entire desktop as a result. Gnome (the interface nazis of the linux world) attempted to do the same with Gnome 3.x, and that caused alot of fallout (Ive alwasy been an openbox user, so this didnt really affect me much).

From a point of focus, I think that what works is what needs to stay, and what doesnt work needs to be fixed. But then thats an opinion coming from an old war-torn Aspie network security engineer, and probably doesnt mean much to anyone.

Asia stocks up on Japan optimism; Europe opens up

BEIJING (AP) ? Asian and European stock markets rose Thursday, while and Japan's benchmark index hit its highest level in more than a year optimism a new government in Japan will stimulate the country's sluggish economy.

Oil gained in Asian trading to stay above $91 a barrel as markets in Hong Kong and Australia reopened after a two-day Christmas break.

Britain's FTSE 100 rose 0.2 percent to 5,963.21. Germany's DAX added 0.2 percent to 7,651.51. France's CAC-40 rose 0.7 percent to 3,676.60. Wall Street braced for a flat opening, with Dow Jones industrial futures marginally lower at 13,046. S&P 500 futures rose less than 0.1 percent at 1,413.90.

Earlier in Asia, Tokyo's benchmark Nikkei 225 index rose 0.9 percent to close at 10,322.98, its highest finish since March 2011. That added to Wednesday's 1.5 percent gain and took the Nikkei to a 22 percent increase for the year. Hong Kong's Hang Seng gained 0.4 percent to 22,619.78. South Korea's Kospi added nearly 0.3 percent to 1,987.35.

Incoming Japanese Prime Minister Shinzo Abe has called for more public works spending to reinvigorate the economy. He wants the Bank of Japan to raise its inflation target from 1 to 2 percent to drag the country out of two decades of deflation, or steadily declining prices that have deadened economic activity.

To help exporters, Abe also has urged the central bank to take steps to dampen Japan's yen. A strong currency has hurt big exporters such as Toyota by making Japanese products more expensive overseas.

"The message from Japan is clear at the moment, the incoming government will do everything in its power to weaken the yen and stimulate the economy," Australia's IG Markets said in a report.

Benchmarks in Singapore, Taiwan and Australia also posted gains.

Mainland Chinese shares lost ground, with the Shanghai Composite Index falling 0.6 percent to 2,205.90 while the Shenzhen Composite Index lost 0.8 percent to 862.82. Shares in wine producers and aerospace-related companies led the gains.

"The losses were a technical correction after the recent gains" said Xu Xiaoyu, an analyst at China Investment Securities, based in Beijing. Kweichow Moutai Co., China's largest maker of "baijiu" liquor, gained 1.1 percent. Sichuan Tuopai Shede Wine Co. gained 6 percent following earlier losses.

"Upcoming festivals might boost demand," Xu said.

On Wednesday, U.S. stocks fell for a third session as trading resumed after the Christmas break. Disappointing holiday sales weighed heavy on retail companies and investors worried about the impending "fiscal cliff" ? automatic tax and spending cuts due to take effect if the White House and Congress fail to agree on a budget deal. Economists worry that could push the economy into recession.

Benchmark oil for February delivery rose 13 cents to $91.11 in electronic trading on the New York Mercantile Exchange. The contract jumped $2.37 to finish at $90.98 per barrel in thin post-Christmas trading in New York.

In currencies, the euro rose to $1.3266 from $1.3220 late Wednesday in New York. The dollar gained to 85.74 yen from 85.63 yen.

Fancy bullet points in Word and PowerPoint ? Business ...

Q. How can I create my own custom bullet points?

A. In Word and PowerPoint, you can create bullet points that look like your company?s logo or follow a holiday theme. Here?s how.

  1. Apply bullet point formatting to your list in Word or PowerPoint.
  2. Right-click your list and choose Bullets, then Define New Bullets at the bottom of the fly out menu.
  3. Click the Picture button.
  4. Now, you can choose one of the pictures shown or choose the import button from the lower right corner of the dialog box and find the picture of your choice.

I recommend creating a small picture of your desired image in Paint to make sure the details wouldn?t make a smaller bullet look messy. Then, import that one.

Moshi Monsters Moshlings Theme Park (3DS) Review - Nintendo Life

Goopendous or a Moshi-mess?

Moshi Monsters Moshlings Theme Park is based on the Moshi Monsters website, a popular Neopets-style virtual pet game that lets you adopt a monster, customise its home and play mini-games to collect prizes. The franchise's first video game outing took the monsters to a zoo, and now they're taking a break to vacation at a faraway amusement park. This latest game opens with various Moshi Monsters bailing out of a plane over the Moshlings Theme Park, scattering the little critters everywhere. It's your quest to rebuild the theme park, round up the monsters, and play mediocre digital approximations of carnival games.

The main focus of the game is on rebuilding the titular theme park, over six distinct areas. You play as a different Moshi Monster in each area, and work through a relatively linear chain of Arcade games (carnival-style mini-games) and Rescue Rooms in order to eventually unlock that level's Ride (a slightly more ambitious mini-game). The Arcade games include a Breakout clone, Moshi-themed Whack-a-Mole, a claw-prize game, a pie-flinging shooting gallery, and air hockey. None of them are any more exciting than they sound, unfortunately, though the Breakout clone and air hockey can be fun in short bursts.

The Rescue Rooms are where the game's star creatures are released from their oddly similar-looking prisons, and these are more interesting than the Arcade games, but marred by slow movement and slightly sloppy controls. You tap a button to "call" the monsters towards safety on the left side of a level, while manipulating a grabber and boxing glove arm to clear their way of obstacles and pick up gems and batteries. The touch screen controls are wonky here: sometimes a hard left will register as a right before fixing itself, and everything seems to need to happen "on the beat", which can feel unresponsive. Also, having to hit the "call" button to keep your Moshi Monster moving leftward makes for a lot of awkwardly incessant tapping.

Once you rescue your Moshi Monsters or Moshi eggs, you can play with them in the Monster Habitats and Nurseries. You can drag toys into the playroom or feed them treats, and play little stylus-based mini-games to give them a bath, prepare hot dogs for them (as you do), serenade them with Simon Says xylophone melodies, and help them get through exercise courses. The mini-games are simple, and reminiscent of Flash-based website diversions, though the lack of any customisation makes them a step down from the actual Moshi Monsters website games. Younger players will likely enjoy these ? though nearly every activity ends with a prompt to blow into the microphone, so be prepared for some spittle on little 3DS screens.

The final parts of each area are the Ride Regenerator and the subsequent Ride. The Regenerators are simple puzzles, while the Rides are unique mini-games for every level, including rollercoaster runs and and white water rapid rides, among others. These have the most potential of any of the mini-games, but sadly, the controls are a real let down. The first area's ride (a downhill kart ride through a tubular chute) is nigh on uncontrollable, with the touch screen's joystick nowhere near sensitive or responsive enough to gather collectables or avoid obstacles with any precision.

Each of the park's six areas feature these same attraction types, and while some are better than others, the uneven quality only exacerbates one of Moshlings Theme Park's worst problems: nothing is skippable. To unlock the Rescue Rooms in each stage, you need to play the same five Arcade games every time. To unlock the Rides, you have to collect every single battery from each of the Rescue Rooms. The only optional areas are the Nurseries and Habitats ? and these are ironically the most enjoyable parts of the game. For a game aimed at younger players, the insistence on beating every game type and finding every last collectable before advancing stages seems like a real misstep.

The biggest issue, however, and one that will be a constant annoyance to kids and adults alike, is that Moshi Monsters is an exceedingly slow game. Not only in terms of pacing, but movement as well. You move your Moshling around the theme park by tapping left or right on the touch screen, but your monsters move with all the speed of Moshi-molasses. It takes an incredibly long time to get anywhere, and your Moshling will insist on stopping at all points en route. There are no express routes in Moshlings Land. To saunter from one end of an area to the other takes about 70 seconds (yes, we counted!) ? and that's something you have to do routinely to select from all the different activities in each level, as the side-scrolling areas essentially act as animated menus. It's pointless, tiresome and excruciating enough to make you think twice about traipsing back to the beginning of an area for anything.

Moshi Monsters' presentation is nothing to write home about, though the bright colours and cartoony graphics will probably be fine for the game's younger audience, who are used to seeing these creatures in website form. It's essentially an average looking DS game, and the 3D effect only serves to create depth between the screen glass and the still-flat-looking game. The music is mostly looping, low-sample-rate MIDI guitar rock, with a few catchier tunes popping up from time to time.

There are a few thoughtful touches that younger games will appreciate, however, like the tactile opening of presents by sweeping the stylus back and forth across the on-screen package. Better still is the game's use of the 3DS' features: there's StreetPass integration for swapping high scores and exchanging "Visitors" (monsters that wander around the park and can come with bonuses), and a cool little AR mini-game (using the '?' card included with the 3DS) to unlock bonus Moshlings. These extras don't make up for the game's problems, but they're welcome additions nonetheless.

Sometimes licensed games made for younger players can surprise us, and exceed our expectations. Sadly, this is not one of those times. Moshlings Theme Park eschews the customisation-based gameplay of its source material in favour of generic, forgettable arcade games, cutting out much of what makes the franchise popular in the process. Poor controls and slow movement pull it down even further, and its strictly average presentation does nothing to help. The game comes with a coupon for a seven day Premium Membership to the Moshi Monsters website in the box, and we'd bet that most players would be much happier with that offer than the actual game.

Ranch And Land For Sale in Valparaiso IN

If you are planning to acquire a Farm and Ranch in Valparaiso IN, you ought to settle on lots of vital things prior to making the acquisition. Initially, you need to choose the county in Valparaiso IN where you want to get a farm and ranch. Whether it will be Jasper, LaPorte, Lake, Porter or Newton ? it all depends on you. Decide on the county relying on the type of farm you wish to develop.

Setting up a Farm and Ranch in Valparaiso IN is fairly a strenuous project particularly if you do not have any sort of prior experience or expertise connected to farm building. Prior to jumping on to this undertaking, it is necessary to decide the sort of farm and ranch you wish to create. Pet ranches, bird farm, dairy and poultry farm, or a farm that produces vegetables and fruits? you need to choose the kind of farm prior to advancing on the next actions. Due to the fact that each kind has its very own requirements, hence the land of the farm should be selected according to the kind you wish to build. When you have actually made that choice, it is time for you to move forward.

Locate an Experienced Realtor to Get a Farm and Ranch in Valparaiso IN

To develop a Farm and Ranch in Valparaiso IN, you would require the solutions of a high-class Farm or Rural Land real estate broker. Usually, realtors have a best understanding of urban property and houses; there are few realtors whose expertise lies in farming and rural areas. Get in touch with an agent of that sort: the one who is well-informed about farms and rural real estate. Discuss your demands and budget with your realtor so he can show you lands as per your needs. Ask him to show you a minimum of eight or ten excellent areas so you have an assortment to choose from. Never ever settle on the first rural real estate you visit. It is most effectively to keep your options open as you may discover a land best and much more cost effective than the first one. Hence, be very sensible in making that choice. You will be investing a handsome quantity of cash in the land so you have to be really sure before acquiring the farmland. Ask your real estate agent to show you currently installed farms in addition to home for constructing a farm from scratch. If you are able to find an established farm at a fair rate, it will be smarter to go through that bargain.

Assess the Location of Farm and Ranch in Valparaiso IN

Whenever you are shown some home to construct a ranch, review it minutely and see that you know every nitty-gritty of it. You should discover about all the farmer?s markets close to that farm location. If your circulation networks and agriculturalist?s market are found at a great distance from that area, it will not be that useful for you. Even if you are being provided a great rate for that Farm and Ranch in Valparaiso IN, you will not be conserving much in the long run, as your fuel and transport costs will certainly be too high. For that reason, it is a good thing to have a ranch close to your providers and markets. This will not just conserve your cash but additionally will certainly curtail the wastage of time.

An additional essential thing you have to evaluate is the land history where you want to create your ranch. Ask the seller to give every information of that area. Typically, sellers have the tendency to hold back any adverse details so it will be best if you question a few of the surrounding ranches in that location. CNN Charles Durning

By Tom Cohen

?

(CNN) ? You may not know his name, but anyone who watches movies and television knows the squat, tough-faced character actor Charles Durning, who died on Christmas Eve, according to his family.

?

The 89-year-old decorated World War II veteran had an award-winning career that spanned 20th Century entertainment, from the burlesque stage and theater to television and film, playing roles as diverse as society itself.

Durning was the gruff father smitten by Dustin Hoffman?s female impersonation of ?Tootsie,? the Southern politician dancing on stage with George Clooney?s Soggy Bottom Boys in ?O Brother, Where Art Thou?? and the corrupt cop caught up in the ?big con? by Paul Newman and Robert Redford in ?The Sting.?

He won Tony and Golden Globe awards and was nominated for Academy Awards two times as a supporting actor. He also received nine Emmy nominations and was honored in 2007 with a lifetime achievement award from the Screen Actors Guild.

Born in Highland Falls, New York, on February 28, 1923, Durning became an actor relatively late.

According to his TV Guide biography, Durning left home at age 16 and later enlisted in the Army. Durning landed on Normandy beach on D-Day and fought in the Battle of the Bulge, receiving the Silver Star and three Purple Hearts.

He worked odd jobs that ranged from professional boxing to teaching ballroom dancing to ushering in a burlesque house, where he got his start on stage by once filling in for a comic who failed to show up, the actor often recalled.

He also attended the American Academy of Dramatic Arts, where he trained with Jason Robards, Colleen Dewhurst and others.

Some of Durning?s other best-known roles included his Tony-winning performance as Big Daddy in ?Cat on a Hot Tin Roof?, a complicit sheriff in ?Best Little Whorehouse in Texas?, a beleaguered police officer in ?Dog Day Afternoon? and as Doc Hopper, the owner of a frog leg restaurant in ?The Muppet Movie.?

Durning?s family, in a statement issued Tuesday, said, ?Not only was Charlie a World War II hero but he was also a hero to his family. Charlie loved Christmas and if he could have chosen a time to pass, he would have chosen this day.?

Scientists sequence genome of pathogen responsible for pneumocystis pneumonia

Dec. 26, 2012 ? Scientists have sequenced the genome of the fungus Pneumocystis jirovecii, an advancement that could help identify new targets for drugs to treat and prevent Pneumocystis pneumonia, a common and often deadly infection in immunocompromised patients. The study will be published on December 26, 2012 in mBio?, the online open-access journal of the American Society for Microbiology. The organism cannot yet be isolated and grown for study in the laboratory, so details about Pneumocystis pneumonia, the biology of P. jirovecii, and its pathogenicity are hard to come by. The genome sequence represents a wealth of new information for doctors and researchers tackling this disease.

Pneumocystis pneumonia is an opportunistic infection that strikes most often in individuals with diminished immune systems. The corresponding author of the study in mBio?, Philippe Hauser of the Centre Hospitalier Universitaire Vaudois and University of Lausanne, in Switzerland, says the disease gained importance in the 1980s.

"Recognized first among malnourished infants, P. jirovecii pneumonia became a public issue with the advent of the HIV epidemic," says Hauser. Today, the disease most commonly affects HIV-infected persons who are unaware of their status as well as solid organ transplant recipients and patients with hemato-oncologic or autoimmune diseases. Since the organism cannot be grown in the lab for study, researchers have long made do with studying P. jirovecii's lab-friendly relatives, species that infect animals and plants, in order to explore the secrets of the human disease.

"It is obviously better to study [P. jirovecii's] genes rather that those of Pneumocystis species from animal models. The genome has both medical and evolutionary interests for the scientific community," says Hauser.

Under normal circumstances, scientists sequencing the genome of a microorganism simply extract DNA from thick cultures of cells they grow in the lab. Since they were unable to grow P. jirovecii cells for their genomic DNA, Hauser and his colleagues took a different approach. They took a sample of bronchoalveolar lavage fluid from an individual infected with Pneumocystis pneumonia, then concentrated the P. jirovecii cells using immuno-precipitation and created copies of the DNA in the sample using a technique called random DNA amplification. This mixture of DNA strands, from P. jirovecii, human, and other microbes from the lungs of the infected patient, was then sequenced using high throughput technologies.

According to Hauser and his colleagues, the fact that the sequence data represented DNA from many different species created the biggest challenge they faced. "The major challenge of the study was the in silico sorting of the reads out of a mixture representing the human host and different organisms present in the lung microbiome," he says. This challenge was met through a collaboration with Marco Pagni of the Vital-IT group of the SIB Swiss Institute of Bioinformatics, who provided indispensable expertise and infrastructure.

Once the sorting task was accomplished, the researchers assembled the sequences into a genome and attempted to identify the functions of P. jirovecii's genes. This is the first time scientists have assembled the genome of a fungus from a mixed pool of DNA from a single source, often called a metagenome. Their analyses reveal a surprising fact: P. jirovecii is a parasite that must live within the human body to survive.

P. jirovecii lacks the genes necessary for creating some of the essential ingredients of life, a hallmark of obligate parasites, organisms that must rely on another creature for sustenance. "It implies that they need their host to provide these molecules. Thus, this has been quite an important finding which implied that human beings represent the reservoir of this pathogen," says Hauser. This is useful information, since it means that people are the only significant source of the organism and that both infected people and healthy carriers represent the only control points for limiting the spread of the disease.

The genome also shows that P. jirovecii apparently lacks the ability to make toxins and virulence factors, molecules that enable a microbe to invade and take advantage of its host. This makes sense, since P. jirovecii does not cause disease in healthy people, but only runs out of control when it is not confronted with an immune response.

In the study of infectious disease, access to the genome of a pathogen provides new information that can be pivotal in combating the diseases is causes. The hope is that the genome of P. jirovecii will lead to new advances in therapies for those suffering from Pneumocystis pneumonia. The current drugs of choice for treating Pneumocystis pneumonia are antifolates, but certain isolates of P. jirovecii have already developed resistance to antifolates, an ability that is very likely to spread. Now that the genome of P. jirovecii is assembled and available to researchers all over the world, scientists can tease out clues about the organism that will help identify targets for some badly needed new drugs.
